I suggest you to take a bus from Firenze to Siena:
Specifically from “Firenze Autostazione” near SMN, to “Siena Viale Tozzi” near your destination.MonicaSpettatoreHi Jenny.
From Positano to Amalfi you need the line 5070:
There’s no direct line from Positano to Naples. You will have to change in Sorrento. There you can take a bus or circumvesuviana (both EAV) to Naples:
In April-May the bus lines could be expanded.

To reach San Gimignano from Siena you need the line 130/A (130/R for the return trip).For Thursday or Friday you have to watch: “GIO” “VEN” “Feriale escluso il sabato” e “Tutti i giorni feriali”.

I do not recommend a one day trip for all destinations. To reach San Gimignano and Monteriggioni you will have to change bus in Poggibonsi, from the same place where you will arrive.For timetables, next links and attach.
